During an infection in humans by any pathogenic microorganism, different classes of antibodies arise. Among these antibodies are adhesins. An adhesin when combined with the homologue antigen (adhesinogen) under controlled conditions can cause adhesion.

Rose Bengal serotype uses Brucella microbial antigen for early detection of adhesins against species of the genus Brucella (Brucella abortus, Brucella melitensis and Brucella suis). This test is an initial screening test only for the detection of adhesins against Brucella. If the result is positive, it should be confirmed by other more specific serological tests for brucellosis.

Detectable adhesins do not always indicate bacterial infection. In some cases, nonspecific adhesins may be present which react with Brucella antigens. For example, sera from certain classes of drug addicts have been reported to contain significant amounts of adhesins. In some cases, cross-reactions may occur, such as tularemia. In addition, vaccinations can produce adhesins capable of reacting with soluble antigens.
